Overview

Bepranemab is a recombinant humanized IgG4 monoclonal antibody developed as an immunotherapy targeting tau protein. It specifically binds to a mid-region epitope of human tau (amino acids 235–250 near the microtubule-binding domain), which is thought to be essential for tau aggregation and propagation in neurodegenerative diseases. By binding this region, bepranemab aims to inhibit the cell-to-cell spread of pathogenic aggregated tau, potentially slowing disease progression. The drug is being investigated primarily for progressive supranuclear palsy (PSP) and Alzheimer's disease. In clinical trials, it has shown the ability to slow accumulation of pathological tau in the brain and may offer clinical benefit in subgroups with low baseline tau or without ApoE4 genetic risk factors[1][2][3][4][5][8]. Bepranemab is administered intravenously as a solution for infusion[1].
