Berahyaluronidase alfa + pembrolizumab is an investigational subcutaneous formulation combining pembrolizumab (an anti-PD-1 therapy) with berahyaluronidase alfa (a hyaluronidase variant). This combination allows for subcutaneous administration of pembrolizumab rather than intravenous infusion. Berahyaluronidase alfa temporarily degrades hyaluronic acid in the extracellular matrix, facilitating the absorption of pembrolizumab when administered subcutaneously. Phase 3 trials have demonstrated noninferior pharmacokinetics compared to IV pembrolizumab, with a median injection time of just two minutes, resulting in nearly 50% reductions in patient chair and treatment room time.
