Overview

Berbamine is a bisbenzylisoquinoline alkaloid isolated from plants such as Cocculus orbiculatus. It is classified as a small molecule and has been studied for its potential pharmacological activities including anti-tumor effects (notably in KRAS-mutant lung adenocarcinoma), calcium channel blocking activity (inhibiting calcium-induced contraction of smooth muscle), and possible neuroprotective properties. Mechanistically, it acts as a calcium channel blocker and has been shown to modulate the STING pathway in cancer cells by downregulating p-STING (Ser366) and CCL2 expression. This leads to reduced infiltration of myeloid-derived suppressor cells into the tumor microenvironment and enhances anti-tumor immunity. Berbamine also exhibits pro-apoptotic effects by triggering cell cycle arrest and promoting cellular senescence in cancer models[7][8][10]. It is considered investigational with ongoing research into its therapeutic applications.
