Overview
Berberine + danazol is an investigational combination therapy being studied for the treatment of corticosteroid-resistant or relapsed immune thrombocytopenia (ITP). Berberine is a plant-derived isoquinoline alkaloid with multiple pharmacological activities, including modulation of gut microbiota, anti-inflammatory effects, and metabolic regulation. It may help correct corticosteroid resistance in ITP by altering gut microbiota structure. Danazol is a synthetic attenuated androgen that inhibits pituitary gonadotropin secretion and has been used as a second-line agent in ITP due to its immunomodulatory properties. The rationale for combining these agents lies in their disparate mechanisms of action, aiming to maximize efficacy while minimizing adverse effects[2][3][8].
