Overview
Bersacapavir (JNJ-56136379, also known as JNJ-6379) is an orally administered small molecule drug developed as a capsid assembly modulator for the treatment of chronic hepatitis B virus (HBV) infection. It acts by binding to the HBV core protein and disrupts viral replication through two main mechanisms. The primary mechanism accelerates capsid assembly kinetics, preventing encapsidation of polymerase-bound pregenomic RNA and leading to the formation of empty capsids—a late-stage process in the HBV life cycle. The secondary mechanism inhibits de novo formation of covalently closed circular DNA (cccDNA), interfering with early-stage processes in viral replication. Bersacapavir has demonstrated potent antiviral activity in vitro and has been evaluated in phase 1 and phase 2 clinical trials for chronic hepatitis B[1][3][6][7].
