Berzosertib + gemcitabine is a combination therapy under investigation for the treatment of advanced solid tumors, particularly platinum-resistant high-grade serous ovarian cancer. Berzosertib (formerly known as M6620 or VX-970) is a first-in-class, highly potent and selective inhibitor of ataxia telangiectasia and Rad3-related protein kinase (ATR), a key regulator in the DNA damage response pathway. Gemcitabine is a nucleoside analog used as chemotherapy that inhibits DNA synthesis by incorporating into DNA and causing chain termination. The rationale for combining these agents stems from preclinical evidence showing synergistic antitumor effects when ATR inhibition follows DNA-damaging chemotherapy like gemcitabine; this schedule exploits increased ATR activity during S-phase arrest induced by gemcitabine. Clinical trials have demonstrated that this combination can improve progression-free survival compared to gemcitabine alone in patients with recurrent or resistant ovarian cancers[1][2][8].
