Overview
Besifovir dipivoxil maleate is an oral small molecule prodrug developed for the treatment of chronic hepatitis B virus (HBV) infection. It is a nucleotide analog that acts as an inhibitor of HBV polymerase by competitively binding to the viral polymerase and terminating DNA chain elongation during viral replication. Besifovir dipivoxil maleate demonstrates potent antiviral activity with a high barrier to resistance and has shown efficacy comparable to tenofovir disoproxil fumarate (TDF), but with improved renal and bone safety profiles. The drug was approved in South Korea in 2017 for chronic hepatitis B. Clinical studies have demonstrated its long-term efficacy and low toxicity over several years of use[1][2][3][5][6].
