Beta 1 integrin shRNA is an RNA interference (RNAi) therapeutic candidate designed to silence the expression of the beta 1 integrin subunit (ITGB1). Integrins are a family of heterodimeric transmembrane receptors that mediate cell-extracellular matrix (ECM) interactions, which are often upregulated in pancreatic cancer and contribute to tumor progression, migration, and metastasis. This specific shRNA is delivered via a lentiviral vector to target ITGB1 mRNA, leading to a significant reduction in protein expression. Preclinical research has shown that knockdown of the beta 1 integrin subunit inhibits cell adhesion, proliferation, and migration on various ECM proteins such as collagen and fibronectin. In orthotopic mouse models of pancreatic cancer, treatment with beta 1 integrin shRNA has been shown to significantly inhibit tumor growth and completely prevent metastasis and ascites formation, highlighting its potential as a targeted therapy for aggressive solid tumors.
