Overview

beta-arrestin2 siRNA is a small interfering RNA (siRNA) designed to silence the expression of the *ARRB2* gene, which encodes the beta-arrestin 2 protein. Beta-arrestin 2 is a multifunctional adaptor protein traditionally known for its role in the desensitization and internalization of G-protein-coupled receptors (GPCRs). In the context of oncology, specifically multiple myeloma (MM), beta-arrestin 2 has been identified as a potential therapeutic target due to its correlation with poor clinical outcomes and resistance to proteasome inhibitors like bortezomib. Preclinical research, such as that presented at ASH 2024, indicates that knockdown of beta-arrestin 2 using siRNA can down-regulate the expression of immune checkpoints, including PD-1 in T cells and PD-L1 in myeloid-derived suppressor cells (MDSCs), thereby potentially enhancing anti-tumor immune responses within the bone marrow microenvironment.
