Overview

beta-d-n4-hydroxycytidine is a **small-molecule ribonucleoside analog antiviral** that acts as the active antiviral moiety of molnupiravir after prodrug conversion. It is also known as EIDD-1931 and N4-hydroxycytidine. After intracellular phosphorylation to its 5-triphosphate metabolite, it serves as a competitive substrate for viral RNA-dependent RNA polymerase and is incorporated into nascent viral RNA, where ambiguous base pairing drives transition mutations and ultimately viral error catastrophe. The compound has shown broad preclinical antiviral activity against multiple RNA viruses including coronaviruses, influenza, chikungunya, Ebola, hepatitis C, MHV, and MERS-CoV, and has been principally associated with development programs originating from Emory-affiliated discovery efforts and later advanced clinically through its oral prodrug molnupiravir for COVID-19.
