Overview

beta-Escin is an oleanane-type pentacyclic triterpenoid saponin extracted from the seeds of horse chestnut (Aesculus hippocastanum). It is primarily used as an anti-inflammatory and vasoconstrictive agent, indicated for the treatment of minor muscle and joint trauma, chronic venous insufficiency, hemorrhoids, inflammation, and edema. Its pharmacological effects include anti-edematous, anti-inflammatory, venotonic, antitumor, and antiviral activities. Mechanistically, beta-Escin inhibits Nuclear factor kappa-light-chain-enhancer of activated B cells (NF-kappaB) activation in endothelial cells under inflammatory conditions—leading to reduced migration and permeability—and modulates several signaling pathways such as ATM/H2AX, RhoA/Rock, GSK3/beta-catenin, HER2/HER3/Akt and PI3K/Akt. It also downregulates Lysyl oxidase homolog 2 (LOXL2) expression (implicated in cancer metastasis) and inhibits Pancreatic lipase. The compound has been shown to protect endothelial integrity against TNF-alpha-induced permeability changes[1][5][6][7][8].
