Overview

**Beta2 adrenergic receptor siRNA** is a small interfering RNA molecule engineered to silence the expression of the beta2 adrenergic receptor (ADRB2 gene) via RNA interference. By specifically targeting and degrading ADRB2 mRNA, it prevents translation of the beta2 adrenergic receptor protein, thereby functionally reducing receptor abundance on cell surfaces. Mechanistically, this diminishes the physiological actions mediated by beta2 adrenergic signaling, most notably in the regulation of smooth muscle tone and aqueous humor dynamics. Preclinical studies in rabbits have shown that topical administration of beta2 adrenergic receptor siRNA results in a robust and long-lasting decrease in intraocular pressure (IOP) (up to 30% reduction), comparable to or longer-lasting than conventional commercial glaucoma medications. No significant side effects on ocular structures have been identified. The main therapeutic interest is for glaucoma, as lowering IOP is the only proven treatment strategy for this disease[1][2].
