Betamethasone microsphere is a sustained-release formulation of the synthetic glucocorticoid corticosteroid betamethasone, designed for prolonged local delivery. The drug utilizes biodegradable polymer-based microspheres to provide controlled release of betamethasone at the site of administration. Its primary mechanism is anti-inflammatory and immunosuppressive action via activation of the glucocorticoid receptor, leading to suppression of inflammatory mediators and immune responses. Betamethasone microsphere (DE-102) has been developed primarily for ophthalmic indications such as diabetic macular edema and other forms of macular edema[1][2][3]. It has also been investigated for transtympanic delivery in inner ear disorders such as Meniere's disease, sensorineural hearing loss, autoimmune inner ear disease, and tinnitus[4]. The sustained-release profile aims to reduce dosing frequency while maintaining therapeutic levels locally.
