Overview

This is a combination therapy consisting of three drugs—bevacizumab, gefitinib, and icotinib—used in the treatment of non-small cell lung cancer (NSCLC), particularly in patients with activating EGFR mutations. Bevacizumab is a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), thereby blocking angiogenesis. Gefitinib and icotinib are both small molecule tyrosine kinase inhibitors (TKIs) targeting the epidermal growth factor receptor (EGFR). The rationale for combining these agents is to simultaneously inhibit tumor cell proliferation via EGFR blockade and suppress tumor angiogenesis via VEGF inhibition, potentially overcoming resistance mechanisms seen with monotherapy. This combination has been explored in clinical settings for advanced or locally advanced NSCLC, especially where EGFR mutations are present[1][3][4][7].
