Overview

This is a combination regimen consisting of three drugs: bevacizumab, a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A); capecitabine, an oral prodrug of 5-fluorouracil that inhibits DNA synthesis; and erlotinib, a small molecule inhibitor of the epidermal growth factor receptor (EGFR) tyrosine kinase. The combination is under investigation for the treatment of advanced cancers, particularly pancreatic cancer. Bevacizumab inhibits angiogenesis by blocking VEGF-A, thereby reducing tumor blood supply. Capecitabine interferes with DNA synthesis in rapidly dividing cells. Erlotinib blocks EGFR signaling pathways involved in cell proliferation and survival. Together, these agents target both tumor cell proliferation and the supporting vasculature[1][2].
