Overview

This is a four-drug combination regimen consisting of bevacizumab, carboplatin, cetuximab, and paclitaxel. It has been studied primarily as a first-line treatment for advanced non-small cell lung cancer (NSCLC), particularly in patients with nonsquamous histology. The regimen combines two monoclonal antibodies—bevacizumab (an anti-VEGF agent) and cetuximab (an anti-EGFR agent)—with two cytotoxic chemotherapeutic agents—carboplatin (a platinum-based DNA crosslinker) and paclitaxel (a microtubule stabilizer). The rationale is to target both tumor angiogenesis and epidermal growth factor receptor signaling while delivering standard chemotherapy. Clinical trials have shown this combination to be feasible, with manageable toxicity profiles and promising efficacy outcomes such as response rate, progression-free survival, and overall survival in NSCLC[1][2][3][4]. Maintenance therapy with bevacizumab and cetuximab may follow initial induction cycles.
