The combination of bevacizumab + cediranib represents a dual inhibition approach to the vascular endothelial growth factor (VEGF) pathway. This combination has been studied in clinical trials for various advanced solid tumors. Bevacizumab is an anti-VEGF-A monoclonal antibody that works by binding to VEGF-A, preventing it from interacting with its receptors. Cediranib (also known as AZD2171 or RECENTIN™) is a highly potent oral tyrosine kinase inhibitor that targets all three VEGF receptors. The rationale behind this combination is to achieve more complete inhibition of the VEGF pathway through complementary mechanisms: bevacizumab targets the VEGF-A ligand directly, while cediranib inhibits the tyrosine kinase activity of VEGF receptors. This dual approach to angiogenesis inhibition may provide enhanced anti-tumor activity compared to either agent alone.
