Bevacizumab-Weill Medical College of Cornell University-glioma refers to a specific investigational treatment protocol for pediatric and young adult patients with relapsed or refractory intracranial gliomas. This therapy combines bevacizumab (Avastin), a monoclonal antibody targeting vascular endothelial growth factor (VEGF) to inhibit angiogenesis, with cetuximab (Erbitux), a monoclonal antibody targeting the epidermal growth factor receptor (EGFR). The treatment is administered using a specialized delivery method called super-selective intra-arterial cerebral infusion (SIACI), where the drugs are infused directly into the arteries feeding the tumor following blood-brain barrier disruption with mannitol. This approach, developed by researchers at Weill Medical College of Cornell University, aims to bypass the blood-brain barrier and achieve high local drug concentrations while reducing systemic side effects. It has been evaluated in Phase I/II clinical trials for high-grade gliomas and diffuse intrinsic pontine glioma (DIPG).
