Bevacizumab is a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), thereby blocking angiogenesis which tumors need for growth. Fluorodeoxyuridine (commonly known as fluorouracil or 5-FU) is a pyrimidine analog that inhibits thymidylate synthase, disrupting DNA synthesis in cancer cells. Oxaliplatin is a platinum-based chemotherapeutic agent that causes DNA crosslinking and apoptosis. The combination of these three agents is used primarily in metastatic colorectal cancer to improve progression-free survival and overall survival compared to chemotherapy alone. Bevacizumab enhances the efficacy of oxaliplatin-based chemotherapy regimens such as FOLFOX by inhibiting tumor blood vessel formation, thus starving tumors of nutrients.
