Overview

This combination therapy consists of bevacizumab, a humanized monoclonal antibody targeting vascular endothelial growth factor (VEGF), and fosbretabulin (CA4P), a vascular disrupting agent (VDA). Bevacizumab acts as an anti-angiogenic agent by neutralizing VEGF-A, thereby preventing the formation of new tumor blood vessels. Fosbretabulin, a prodrug of combretastatin A-4, targets existing tumor vasculature by binding to the colchicine site of beta-tubulin, leading to microtubule depolymerization and the rapid collapse of established tumor vessels. The combination is being evaluated for its potential synergistic effects in recurrent high-grade gliomas, where the anti-angiogenic properties of bevacizumab may complement the vascular-disrupting activity of fosbretabulin to improve clinical outcomes.
