bevacizumab + levofolinic acid + 5-fluorouracil + oxaliplatin is a multi-agent chemotherapy regimen that combines a monoclonal antibody (bevacizumab) targeting vascular endothelial growth factor (VEGF) with the FOLFOX regimen, which includes oxaliplatin (a platinum-based alkylating agent), 5-fluorouracil (an antimetabolite pyrimidine analog), and levofolinic acid (folinic acid, also known as leucovorin, used to enhance the efficacy and reduce toxicity of 5-fluorouracil)[1][2][5]. This regimen is primarily indicated for the first-line treatment of metastatic colorectal cancer (mCRC)[1][2][4]. Bevacizumab inhibits angiogenesis by binding VEGF-A, thus suppressing blood vessel growth in tumors. 5-fluorouracil inhibits thymidylate synthase to block DNA synthesis. Levofolinic acid enhances the binding of 5-fluorouracil to thymidylate synthase. Oxaliplatin induces DNA cross-linking and apoptosis in cancer cells[1][2][5].
