Bevacizumab + marizomib is an investigational combination therapy evaluated primarily for the treatment of recurrent or relapsed WHO Grade IV malignant glioma (glioblastoma). Bevacizumab is a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), thereby blocking angiogenesis and reducing tumor blood supply. Marizomib is an irreversible pan-proteasome inhibitor that crosses the blood–brain barrier and disrupts protein degradation pathways essential for tumor cell survival. The combination aims to target both tumor vasculature and proteasome-dependent cellular processes in glioblastoma cells. Clinical trials have shown that this regimen has a safety profile consistent with each agent individually but did not demonstrate a significant improvement in efficacy over standard therapies for recurrent glioblastoma[1][2][3][7].
