Overview

This is a combination regimen consisting of three drugs—bevacizumab, paclitaxel, and pemetrexed—used primarily in the treatment of advanced non-squamous non-small cell lung cancer (NSCLC). - **Bevacizumab** is a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), thereby blocking angiogenesis and reducing blood supply to tumors. - **Paclitaxel** is a small molecule chemotherapeutic agent that stabilizes microtubules, inhibiting cell division and promoting apoptosis in rapidly dividing cells. - **Pemetrexed** is an antifolate antimetabolite that inhibits several enzymes involved in folate metabolism and DNA synthesis, including thymidylate synthase, dihydrofolate reductase, and glycinamide ribonucleotide formyltransferase. This combination has been studied as part of induction therapy for advanced NSCLC. In clinical trials such as ECOG-ACRIN 5508 and PointBreak, patients typically receive carboplatin with this triplet for up to four cycles; those without progression may continue with maintenance therapy using some components of the regimen[1][4][6]. The triplet has also been explored experimentally for other cancers but its primary use remains in NSCLC[5].
