Overview

Bevacizumab + pazopanib is a combination therapy consisting of two anti-angiogenic agents used primarily in the treatment of advanced or metastatic renal cell carcinoma (RCC). Bevacizumab is a monoclonal antibody that binds to and neutralizes vascular endothelial growth factor A (VEGFA), thereby inhibiting angiogenesis and directly affecting tumor cells by depriving them of nutrients and oxygen. Pazopanib is an oral small molecule tyrosine kinase inhibitor that targets multiple receptors, including VEGF receptors 1, 2, and 3, platelet-derived growth factor receptors (PDGFR) alpha and beta, as well as c-KIT. The combination aims to comprehensively block pathological angiogenesis by targeting both the ligand (VEGFA) with bevacizumab and its receptor signaling with pazopanib. Clinical studies have shown this regimen can significantly improve progression-free survival compared to monotherapy with either agent alone in patients with metastatic clear cell RCC. The combination has demonstrated promising efficacy—doubling median progression-free survival—and an acceptable safety profile in favorable-risk patient populations[1][2][3][4][5][7].
