Overview
The combination of **bevacizumab**, **toripalimab**, and **dacarbazine** is an investigational triplet therapy being evaluated for the treatment of advanced melanoma, particularly mucosal melanoma. **Toripalimab** is a humanized monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, blocking its interaction with PD-L1 and PD-L2 to restore the antitumor activity of T cells. **Bevacizumab** is a recombinant humanized monoclonal antibody that binds to vascular endothelial growth factor (VEGF), inhibiting angiogenesis and potentially normalizing tumor vasculature to improve immune cell infiltration. **Dacarbazine** is an alkylating chemotherapy agent that exerts direct cytotoxic effects by methylating DNA, leading to cell death. This regimen aims to achieve synergistic therapeutic effects by combining immune checkpoint inhibition, anti-angiogenesis, and traditional chemotherapy to overcome resistance in difficult-to-treat melanoma populations.
