A combination drug regimen composed of **bevacizumab** (a monoclonal antibody against vascular endothelial growth factor, VEGF), **trastuzumab** (a monoclonal antibody targeting human epidermal growth factor receptor 2, HER2), and **chemotherapy** (various agents including, but not limited to, docetaxel, paclitaxel, carboplatin, capecitabine, and oxaliplatin). This combination is investigated primarily as a first-line therapy for HER2-positive metastatic cancers, including metastatic breast cancer and metastatic gastroesophageal adenocarcinoma. Bevacizumab inhibits angiogenesis by binding VEGF-A, trastuzumab blocks HER2-mediated signaling, and chemotherapeutic agents exert cytotoxic effects on dividing cells. The regimen has been clinically active, improving response rates and progression-free survival, but generally not overall survival, with increased rates of toxicity.
