Overview

Bevacizumab + valganciclovir is a combination therapy consisting of bevacizumab, a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), and valganciclovir, an oral prodrug of ganciclovir that inhibits viral DNA synthesis. Bevacizumab acts as an anti-angiogenic agent by binding to VEGF-A and preventing its interaction with receptors on the surface of endothelial cells, thereby inhibiting tumor angiogenesis. Valganciclovir is primarily used as an antiviral against cytomegalovirus (CMV) but has been investigated for potential antitumor effects in glioblastoma due to the possible involvement of CMV in tumor biology. The combination has been studied off-label for recurrent glioblastoma multiforme (GBM), where retrospective analyses suggest a trend toward improved survival compared to bevacizumab alone[1][2][3]. However, evidence remains limited and further prospective studies are needed.
