A combination therapy consisting of bevacizumab, a humanized monoclonal IgG1 antibody targeting vascular endothelial growth factor (VEGF), and vorinostat, a small molecule histone deacetylase (HDAC) inhibitor. Bevacizumab works by inhibiting angiogenesis through VEGF blockade, while vorinostat crosses the blood-brain barrier and has anti-tumor effects by inhibiting histone deacetylase and indirectly promoting anti-angiogenic activity. This combination represents a rational therapeutic approach that targets both endothelial cells to block angiogenesis and prevents tumor adaptation to the resulting hypoxia by downregulating angiogenesis-related gene expression.
