Overview

This is a combination chemotherapy regimen consisting of three drugs: bexarotene, carboplatin, and gemcitabine. - **Bexarotene** is a retinoid X receptor (RXR) agonist that modulates gene expression involved in cell differentiation and apoptosis. It is primarily used in the treatment of cutaneous T-cell lymphoma but has been investigated for other malignancies as well. - **Carboplatin** is a platinum-based chemotherapeutic agent that causes DNA crosslinking and inhibits DNA synthesis, leading to apoptosis in rapidly dividing cells. It is commonly used for various solid tumors including lung, ovarian, and bladder cancers[1][2][3][4][5][6]. - **Gemcitabine** is a nucleoside analog (antimetabolite) that inhibits DNA synthesis by incorporation into DNA strands during replication, resulting in cell death. It has broad activity against several solid tumors such as non-small-cell lung cancer (NSCLC), pancreatic cancer, bladder cancer, and others[1][2][3][4][5]. The combination of carboplatin plus gemcitabine (often called "GemCarbo") has established efficacy for advanced NSCLC[2], bladder carcinoma[1], cancers of unknown primary site[5], and other solid tumors; it offers an alternative to cisplatin-based regimens with generally more favorable toxicity profiles[3]. The addition of bexarotene to this doublet would represent an investigational approach aimed at enhancing antitumor effects through complementary mechanisms.
