Bexarotene + carboplatin + paclitaxel is a combination chemotherapy regimen studied primarily for the treatment of advanced non-small cell lung cancer (NSCLC). Bexarotene is a synthetic retinoid and selective retinoid X receptor (RXR) agonist, while carboplatin and paclitaxel are established cytotoxic chemotherapeutic agents. Bexarotene modulates gene expression involved in cell differentiation and apoptosis via RXR activation. Carboplatin is a platinum-based agent that induces DNA crosslinking, leading to apoptosis, and paclitaxel stabilizes microtubules, inhibiting cell division. Clinical studies have shown that this combination is feasible and generally well tolerated; however, it did not significantly improve overall survival or response rates compared to standard chemotherapy alone in NSCLC patients[1][2][7]. The regimen’s main toxicities are consistent with those expected from the individual drugs.
