Bexmarilimab + nivolumab is an investigational combination immunotherapy regimen being evaluated for the treatment of advanced solid tumors, including non-small cell lung cancer (NSCLC) and melanoma. Bexmarilimab is a humanized IgG4 monoclonal antibody that targets Clever-1 (Common Lymphatic Endothelial and Vascular Endothelial Receptor-1), a scavenger receptor expressed on immunosuppressive M2 macrophages. By blocking Clever-1, bexmarilimab aims to reprogram the tumor microenvironment from an immunosuppressive to an immunostimulatory state. Nivolumab is a programmed death-1 (PD-1) immune checkpoint inhibitor that enhances T-cell activity. The combination is being studied in the Phase I/II BLAZE trial (sponsored by the Institute of Cancer Research) to determine if bexmarilimab can resensitize tumors to PD-1 inhibition in patients who have progressed on prior immunotherapy.
