Overview

**BEY1107 + capecitabine** is a combination investigational anti-cancer regimen being studied particularly for **metastatic colorectal cancer refractory or intolerant to standard of care**. BEY1107 (also known as Avotaciclib) is a small molecule, selective inhibitor of cyclin-dependent kinase 1 (**CDK1**), which induces cell cycle arrest at the G2/M phase and promotes apoptosis of cancer cells. It also inhibits AKT signaling, decreases chemoresistance and metastasis factors, and suppresses cancer stem cell self-renewal, broadening its potential antitumor activity. Capecitabine is an oral prodrug of 5-fluorouracil (5-FU), a pyrimidine analog antimetabolite that interferes with DNA synthesis and inhibits cell proliferation. This specific combination is in phase 1/2 clinical trials for metastatic colorectal cancer and is being developed and trialed by BeyondBio. The goal of the combination is to improve response rates and overcome resistance observed with current cytotoxic regimens through dual targeting of cell cycle and nucleic acid metabolism pathways[1][3][7].
