Bezitramide

Bezitramide is a potent, orally active, and long-acting narcotic analgesic that functions as an opiate agonist. It has been utilized for the management of severe chronic pain and postoperative pain. Pharmacokinetic studies indicate oral absorption, with an apparent elimination half-life ranging from 11 to 24 hours. Its active metabolite is despropionyl-bezitramide, and other metabolites include 1-(4-piperidinyl)-1,3-dihydro-2H-benzimidazole-2-one and 3-cyano-3,3-diphenylpropionic acid. More recently, bezitramide has been identified through virtual screening as a potential candidate for drug repurposing to inhibit Epstein-Barr nuclear antigen 1 (EBNA1) for the treatment of Epstein-Barr virus-related malignancies.
