This therapeutic regimen is a multi-agent combination therapy being evaluated by the University of Michigan Rogel Cancer Center for the treatment of adults with Philadelphia Chromosome-Positive (Ph+) Acute Lymphoblastic Leukemia (ALL). The regimen utilizes a pediatric-inspired Berlin-Frankfurt-Münster (BFM) chemotherapy backbone, which includes a diverse array of cytotoxic agents such as daunorubicin, vincristine, pegaspargase, cyclophosphamide, and methotrexate, integrated with the tyrosine kinase inhibitors (TKIs) dasatinib and ponatinib. The Phase II study (NCT03501368) aims to improve clinical outcomes in adult Ph+ ALL patients by combining intensive chemotherapy with continuous TKI administration, alternating between dasatinib and ponatinib to target the BCR-ABL1 fusion protein while managing overlapping toxicities. The regimen also incorporates rituximab for CD20-positive disease and intrathecal chemotherapy for central nervous system prophylaxis.
