BG-C0979 is an investigational antibody-drug conjugate (ADC) developed by BeOne Medicines that targets Protein Tyrosine Kinase 7 (PTK7). PTK7 is a pseudo-kinase receptor that is overexpressed in various solid tumors, including prostate, lung, and esophageal cancers, and is associated with tumor progression and poor prognosis. The ADC consists of a humanized monoclonal antibody against PTK7 conjugated to a cytotoxic payload, typically a topoisomerase I inhibitor, via a cleavable linker. Upon binding to PTK7 on the tumor cell surface, the complex is internalized, leading to the intracellular release of the payload, which induces DNA damage and cell death. BG-C0979 is currently being evaluated in Phase 1 clinical trials as a monotherapy and in combination with the PD-1 inhibitor tislelizumab for patients with advanced solid tumors, including metastatic castration-resistant prostate cancer (mCRPC).
