Drug intelligence / Profile preview
BG-C477 + tislelizumab is an investigational combination regimen consisting of BG-C477, a novel antibody-drug conjugate (ADC) targeting carcinoembryonic antigen-related cell adhesion molecule 5 (CEACAM5/CEA), and tislelizumab, a humanized IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1). Developed by BeOne Medicines (formerly BeiGene), BG-C477 comprises a CEA-specific antibody conjugated to a topoisomerase I inhibitor payload via a cleavable linker, designed to deliver cytotoxic therapy directly to CEA-expressing tumor cells while inducing bystander killing of neighboring tumor cells. Tislelizumab acts as an immune checkpoint inhibitor, blocking the interaction of PD-1 with its ligands PD-L1 and PD-L2 to restore T-cell-mediated antitumor activity. The combination is being evaluated in a Phase 1a/1b clinical trial (NCT06596473) for the treatment of selected advanced solid tumors.
