Overview

BG129 is a novel small molecule inhibitor of Ataxia telangiectasia and Rad3-related (ATR) protein kinase, a key regulator of the DNA damage response (DDR) pathway. Developed by Merrimack Pharmaceuticals, BG129 is designed to be used in combination with DNA-damaging agents, such as carboplatin, gemcitabine, or irinotecan, to enhance their anti-tumor activity by blocking DNA repair mechanisms in cancer cells. To address the rapid clearance observed with early ATR inhibitors, BG129 was formulated into nano-liposomes (nLs-BG129). This formulation significantly improves the pharmacokinetic profile, providing prolonged clearance and enhanced efficacy in preclinical models of ovarian and lung cancers.
