Overview

BG505 MD39.3 gp151 mRNA is an experimental mRNA-based vaccine candidate developed by Moderna in collaboration with the National Institutes of Health (NIH), the International AIDS Vaccine Initiative (IAVI), and Scripps Research. As a component of the mRNA-1574 program, this specific construct encodes a stabilized, native-like HIV-1 envelope (Env) trimer antigen derived from the BG505 strain. The "MD39.3" designation refers to a highly stabilized version of the SOSIP trimer, while "gp151" indicates a specific C-terminal truncation of the glycoprotein designed to optimize the presentation of the trimer to the immune system. The vaccine utilizes Moderna's lipid nanoparticle (LNP) technology to deliver the mRNA, which then directs host cells to produce the Env trimer to elicit autologous and broadly neutralizing antibodies (bNAbs). It is currently being evaluated in Phase 1 clinical trials, such as HVTN 302, to assess safety and immunogenicity in healthy, HIV-uninfected adults.
