Overview

BG505 SOSIP.664 gp140 is a next-generation, cleaved, glycosylated trimeric HIV-1 envelope glycoprotein immunogen designed to mimic the native structure of the HIV-1 Env spike protein[4][2][3]. The construct is based on the subtype A transmitted/founder virus sequence (BG505) and incorporates specific modifications—including an engineered disulfide bond (SOS), an I559P substitution for stability in pre-fusion conformation (IP), and truncation at residue 664—to stabilize the trimer and prevent aggregation[1][3]. The membrane-proximal external region (MPER) is deleted to improve solubility[3]. This recombinant protein displays epitopes for broadly neutralizing antibodies while occluding non-neutralizing antibody sites[3]. It is being developed primarily as a vaccine candidate for HIV infection prevention and has demonstrated immunogenicity in preclinical models as well as safety and immunogenicity in early-phase clinical trials[4][5].
