**BGB-16673 + zanubrutinib** is an investigational oral combination therapy under evaluation for the treatment of patients with relapsed or refractory B-cell malignancies, including non-germinal center B-cell diffuse large B-cell lymphoma, follicular lymphoma, Richter’s Transformation, Waldenstrom macroglobulinemia, marginal zone lymphoma, mantle cell lymphoma, and chronic lymphocytic leukemia/small lymphocytic lymphoma. BGB-16673 is a small molecule Bruton tyrosine kinase (BTK) degrader that targets and promotes degradation of BTK protein, potentially overcoming resistance to traditional BTK inhibitors. Zanubrutinib is a highly selective small molecule inhibitor of Bruton's tyrosine kinase that inhibits B-cell receptor signaling, leading to reduced proliferation and survival of malignant B-cells. Both drugs are administered orally. The combination is being studied for its synergistic potential to produce deeper and more durable responses in B-cell malignancies, especially in patients previously exposed to BTK inhibitors or with relapsed/refractory disease. Developed and manufactured by BeiGene, neither BGB-16673 nor its combination with zanubrutinib have been FDA approved for any indication as of September 2025[1][2][3][5].
