Overview

BGB-24714 is an orally bioavailable small molecule that acts as a second mitochondrial-derived activator of caspases (Smac) mimetic and functions as an antagonist of cellular inhibitor of apoptosis proteins (IAPs), including cIAP1 and XIAP. By inhibiting these IAPs, BGB-24714 promotes apoptosis in cancer cells, potentially enhancing the effects of chemotherapy and radiotherapy. Preclinical studies have shown that it induces degradation of cIAP1, antagonizes the inhibitory interaction between XIAP and caspase-9, leading to caspase activation and tumor cell death. It has demonstrated significant anti-tumor activity both as a monotherapy and in combination with paclitaxel in preclinical models. The drug is being developed by BeiGene for the treatment of advanced or metastatic solid tumors[1][4][5][8].
