BGB-30813 is an orally bioavailable small molecule inhibitor of diacylglycerol kinase zeta (DGKζ), developed for the treatment of advanced or metastatic solid tumors. By inhibiting DGKζ, the drug modulates T cell receptor signaling pathways and may enhance antitumor immune responses. It is currently being evaluated as monotherapy and in combination with the anti-PD-1 monoclonal antibody tislelizumab in Phase 1 clinical trials for a broad range of solid tumors, including non-small cell lung cancer (NSCLC), head and neck squamous cell carcinoma (HNSCC), small cell lung cancer, hepatocellular carcinoma, esophageal cancer, gastric or gastroesophageal carcinoma, nasopharyngeal carcinoma, triple-negative breast cancer, urothelial carcinoma, renal cell carcinoma (RCC), cervical cancer, endometrial carcinoma, cutaneous squamous cell carcinoma, melanoma and Merkel cell carcinoma.
