BGB-45035 is an investigational, orally administered small molecule designed as a highly selective and potent degrader of interleukin-1 receptor-associated kinase 4 (IRAK4) using chimeric degradation activation compound (CDAC) or proteolysis-targeting chimera (PROTAC) technology. By promoting targeted degradation of IRAK4, BGB-45035 aims to completely inhibit IRAK4 function, thereby blocking key inflammatory signaling pathways involved in autoimmune and dermatological diseases. Preclinical studies have demonstrated strong and sustained anti-inflammatory effects with superior efficacy compared to other IRAK4 degraders. The drug is currently being evaluated in first-in-human Phase 1 clinical trials for safety, tolerability, pharmacokinetics, and pharmacodynamics in healthy volunteers as well as adults with autoimmune dermatological conditions such as atopic dermatitis and prurigo nodularis[2][3][5][8].
