BGB-B167 + tislelizumab is an investigational combination therapy comprising **BGB-B167**, a first-in-class IgG-based bispecific antibody targeting carcinoembryonic antigen (CEA) and 4-1BB (CD137), and **tislelizumab**, an anti-PD-1 monoclonal antibody. BGB-B167 is designed to simultaneously bind **CEA** on tumor cells and **4-1BB** on activated T lymphocytes, facilitating T-cell receptor co-stimulation in the presence of tumor antigen, thereby enhancing localized antitumor T-cell activation while aiming to mitigate systemic adverse events. Tislelizumab blocks the PD-1/PD-L1 immune checkpoint pathway, further promoting anti-tumor immunity. This combination is under early-phase clinical investigation (Phase 1a dose escalation, NCT05494762) for patients with advanced or metastatic **CEA-positive solid tumors** (including colorectal cancer (CRC), gastric cancer (GC), and non-small cell lung cancer (NSCLC)) who have progressed after standard therapies. Early results suggest the combination is generally well-tolerated and can confer antitumor responses in select patients[1][3][4].
