Drug intelligence / Profile preview
BGB-B2033 + tislelizumab + bevacizumab is an investigational combination regimen being evaluated for the treatment of advanced or metastatic solid tumors, particularly glypican-3 (GPC3)-expressing malignancies such as hepatocellular carcinoma (HCC). The regimen consists of three active components: BGB-B2033, a bispecific monoclonal antibody that simultaneously targets GPC3 on tumor cells and the costimulatory receptor 4-1BB (CD137) on T cells to drive localized immune activation; tislelizumab, an anti-PD-1 monoclonal antibody designed to inhibit immune checkpoint signaling; and bevacizumab, an anti-VEGF monoclonal antibody that inhibits angiogenesis. By combining tumor-localized 4-1BB costimulation, immune checkpoint blockade, and anti-angiogenesis, this triplet therapy aims to synergistically enhance anti-tumor immune responses and overcome resistance in the tumor microenvironment. It is currently undergoing Phase 1/2 clinical development.
