BGB-C354 + tislelizumab is an investigational **combination therapy** under clinical development for advanced solid tumors. BGB-C354 is an antibody-drug conjugate (ADC) targeting B7-H3 (CD276), a tumor-associated antigen highly expressed in various solid tumors, designed to deliver cytotoxic payloads directly to B7-H3 expressing cancer cells, causing DNA damage and apoptosis. Tislelizumab is a monoclonal antibody that targets PD-1 (programmed death-1) receptor, acting as a checkpoint inhibitor to enhance T-cell-mediated immune responses against tumor cells. The combination aims to leverage BGB-C354’s targeted cytotoxicity together with the immune checkpoint blockade of tislelizumab for potential synergistic antitumor activity. The combination is being studied in a first-in-human Phase 1a/1b clinical trial evaluating safety, tolerability, pharmacokinetics, and preliminary antitumor activity in adults with advanced, metastatic, or unresectable solid tumors whose cancers are not amenable to curative therapy[1][3][5][7][9].
