BGB-R046 is an investigational IL-15 pro-drug designed for cancer immunotherapy. It remains inactive in circulation and is specifically activated by tumor-enriched proteases within the tumor microenvironment, releasing an IL-15 superagonist to stimulate anti-tumor immune responses. The molecule consists of an IL-15Rα-sushi-IL-15 (IL-15 superagonist), a protease-cleavable linker, a masking moiety, and a half-life extension domain. Upon activation at the tumor site, it induces potent immune cell activation with reduced systemic toxicity compared to unmodified IL-15 therapies. BGB-R046 is being developed primarily for advanced or metastatic solid tumors and has demonstrated significant anti-tumor efficacy in preclinical models with favorable pharmacokinetics and safety profiles[1][2][5].
