BGB-R046 + tislelizumab is a combination investigational oncology therapy consisting of **BGB-R046**, a protease-activated **IL-15 pro-drug** (cytokine therapy) designed to deliver localized immune stimulation within the tumor microenvironment, and **tislelizumab** (BGB-A317), a humanized monoclonal antibody targeting **programmed cell death protein 1 (PD-1)**, functioning as an immune checkpoint inhibitor. BGB-R046 is engineered to remain inactive in circulation and be activated by tumor-associated proteases, releasing IL-15Rα-sushi-IL-15 (superagonist) at the tumor site, enhancing **natural killer (NK) cell** and **CD8+ T-cell** expansion. The combination aims to augment anti-tumor immunity while mitigating systemic toxicity of IL-15. Developed for advanced or metastatic immune-sensitive solid tumors, the combination is under investigation in a first-in-human, phase 1 clinical trial[1][3][4][5].
