BGC0222 is an investigational antibody-drug conjugate (ADC) targeting Claudin 18.2 (CLDN18.2), developed by Gao Rui Yao Ye (Beijing) Technology Co., Ltd. (also known as Glory Pharmaceutical). Claudin 18.2 is a tight junction protein that is highly expressed in several types of solid tumors, including gastric, pancreatic, and esophageal cancers, while having restricted expression in normal tissues, making it an attractive therapeutic target. BGC0222 consists of a humanized anti-CLDN18.2 monoclonal antibody conjugated to a potent cytotoxic payload. Upon binding to CLDN18.2 on the surface of cancer cells, the ADC is internalized via endocytosis, and the payload is released intracellularly to induce cell death. BGC0222 is currently being evaluated in Phase 1 clinical trials for the treatment of advanced or metastatic solid tumors.
