Overview

BGC0228 is a long-acting SN-38 prodrug developed by BrightGene Bio-Medical Technology for the treatment of advanced solid tumors. It consists of SN-38, the potent active metabolite of the topoisomerase I inhibitor irinotecan, conjugated to a polymeric carrier. This design aims to optimize the pharmacokinetic profile of SN-38 by extending its half-life and enhancing tumor accumulation through the enhanced permeability and retention (EPR) effect. By providing a sustained release of the active payload, BGC0228 seeks to improve therapeutic efficacy while minimizing the systemic toxicities, such as severe diarrhea and neutropenia, typically associated with conventional irinotecan. It is currently being evaluated in Phase I clinical trials for a variety of solid tumors, including colorectal, pancreatic, and lung cancers.
